Caspr2-antibdies are associated with a spectrum of neurological core symptoms and signs. The mechanisms by which Caspr2-specific Abs mediate pathology [...] Contactin associated protein-like 2 (Caspr2) encephalitis is a rare Antibody (Ab)-mediated autoimmune encephalitides (AE). Caspr2-antibdies are associated with a spectrum of neurological core symptoms and signs. The mechanisms by which Caspr2-specific Abs mediate pathology and how Ab-mediated effector functions translate into clinical syndromes and disease progression are incompletely understood. This master project aims to characterize the anti-Caspr2 antibody repertoires of Caspr2-AE patients. Using a pipeline routinely used in the ATP lab, Caspr2-specific memory B cells from patients will be sorted, cultured, and tested for specific antibody secretion. Sequencing of antibodies heavy and light chains will be performed and obtained antibody repertoires will be analyzed for diversity, V(D)J gene usage, and constant region identity. Representative sequences of anti-Caspr2 antibodies will be synthesized and cloned into expression vectors for in vitro production as recombinant fully human antibodies. Binding to different Caspr2 domains will be assessed by ELISA and affinity for Caspr2 ECD will be analyzed by biolayer interferometry. Motivated candidates with a strong background in immunology and/or neurobiology and at least one significant experience in a research lab including cell culture manipulations are encouraged to apply. Good communication skills in English and teamwork are essential. Possible starting date: January 2025. | |
